HIV-derived viruses are the most employed lentiviral gene transfer vectors. In this system, one or two helper constructs provide gag, pol, tat, rev and some other essential proteins in trans. Exogenous viral glycoproteins, such as the vesicular stomatitis virus glycoprotein (VSV-G), provided by another seperate plasmid, are used as envelop. The transfer vector contains the transgene cassette. The helper and envelop constructs, together with the transfer vector, are co-transfected into HEK293T cells to produce viruses containing a transgene.
